In the last week we have learned how to maintain the stability of genetic material between the parents and offspring, and this week we will study the instability of genetic material caused by DNA mutations. DNA mutations are often spontaneously occur or environmental factors induced. And the mutations spontaneously caused by DNA replication will be addressed in this lecture. We will start with the concept and types of various DNA mutations, and followed by how the mismatch mutations in replication can be repaired in cell.
Key points:
1. Mutation
Ø Base substitution: transition, transversion, point mutation
Ø Base insertion or deletion: frameshift mutation (Not all base insertions or deletions will result in frameshift mutation)
Ø Silent mutation, Missense mutation, Nonsense mutation
2. Mutations caused by replication
Ø Point mutation caused by base mismatch
Ø DNA microsatellite sequences and copy number variations cause by template slips in replication
3. DNA mismatch repair system
Ø The mismatch repair system in E. coli
The functions of MutS dimer, MutL and MutH
The process of mismatch repair
The mechanism to recognize the nascent chain that needs to be repaired instead of parental chain.
