For eukaryotic precursor mRNA, in addition to the removal of intron sequence by splicing, there are another two kinds of modification that are crucial for the maturation of mRNA, the eukaryotic mRNA 5 'end capping and 3' end tailing. These two modifications are necessary for mRNA to be able to transport out of the nucleus effectively and to act as a template for protein translation. What are the cap on the head and the tail behind the body of mRNA and how they are added? This is what we will introduce to you in this section.
The key points of this section include:
1、 5’ capping on eukaryotic mRNA
ü 5' capped eukaryotic mRNA
ü Structure of 5' end cap
ü Type 0, type I and type II cap
ü The process of adding 5' end cap and the enzyme catalyzing each step
ü The 5' capping depends on the CTD phosphorylation of RNA polymerase II
ü Biological significance of 5' end capping
2、 Polyadenylation of eukaryotic mRNA 3' end
ü The function of the factors and enzymes involved in the polyadenylation of 3' terminal (CPSF, CSTF and PAP)
ü The process of polyadenylation at the 3' end (note that its synthesis does not depend on DNA template)
ü The function and significance of the 3' end polyadenylation
