DNA polymerase only proceeds DNA replication from 5′to 3’. As early scientists realized, when the final primer is removed from the lagging strand at the end of a chromosome, this region is left unreplicated. Strict application of these rules to linear chromosomes predicts that chromosomes would get shorter with each round of replication. In this section, you'll understand how eukaryotic cells respond to this problem by telomeres at the ends of chromosomes. Telomeres protect chromosome integrity and prevent non-specific connection of chromosome ends. More importantly, Telomeres are closely related to a variety of physiological and pathological processes such as aging and tumor, so the research on telomeres will be hot for a long time.
The key points of this section include:
1. The problem of linear DNA end replication
2. Characteristics of telomeres
3. The regulation of telomerase
4. The synthesis of two strands of telomere
5. The telomere binding proteins
6. Relationship between the telomere length and the cell division
Please find more literatures on the relationship between telomerase activity and aging/ diseases in our extra online resources if you are interested.
