Key points 7-1 RNA splicing

In this week, we mainly focus on the post transcriptional processing and modification of RNA in eukaryotic cells. A big difference between eukaryotic cells and prokaryotic cells in the process of transcription is that the two processes of transcription and translation are not coupled. After transcription, mRNA needs to undergo a series of processing and modification, such as capping, splicing and tailing. In this section, we will first focus on the characteristics of eukaryotic genes and then introduce the process of RNA splicing. It is believed that the concepts of intron and exon are not new to students, but with the development of life science, their definitions and connotations, especially intron, have changed a lot. There are also some problems that students are easy to confuse, that is, the type II self-splicing intron and the spliceosome spliced intron. They are all the same in structure and splicing mode. The only difference is whether the splicing reaction is catalyzed by the intron sequence itself or by the spliceosome. Hope you may make it clear.

The key points of this section include

1、            The concept of exon and intron

ü  Respective functions and products that can be coded

ü  The quantitative characteristics of introns in different species

ü  Features of intron and exon length

2、            RNA splicing

ü  Splicing is involved in the processing of various eukaryotic RNA

ü  The nomenclature of splice sites and conserved sequences related to splicing (5 'splice sites, 3' splice sites, branch points)

ü  Two steps of transesterification are involved in the splicing of eukaryotic mRNA

3、            Spliceosome mediated splicing of eukaryotic mRNA

ü  Composition of spliceosome U1U2U4U5U6

ü  The function of each snRNP components of spliceosome

ü  Assembly process of spliceosome

4、            Self splicing intron

ü  The splicing process of type I self splicing introns

ü  The splicing process of type II self splicing introns
