The process of transcription is the first step in the flow of genetic information in cells. Even small single cell prokaryotes need to accurately regulate gene expression according to the changes of external environment and their own state. Some genes in the genome need to be continuously expressed, which we call constitutive expression, while some genes need to be induced by external factors before they start to express, which we call induced expression. So these inducible genes are not always transcribed. So, how does a prokaryotic cell control its RNA polymerase to know when and where to begin its work? In this section, we will introduce some basic concepts of transcriptional regulation and the gene expression regulation mechanism of prokaryotic cells, taking the most well studied classical lactose operon as an example.

The operon model:regulator gene、structural genes、operator
Positive control and negative control
Regulatory proteins: activator、repressor
Inducer、Inhibitor

Structure:structural genes:lacZ, lac Y, lac A
regulator gene:lacI
Sequence characteristics of operator and its relationship with lac promoter
Structure of LacI protein and its interaction with operator
The regulatory mechanism of lac operon and its positive control by CAP-cAMP
3. Inhibition of glucose on lactose operon
Catabolite repression:Inducer exclusion
Inhibition of CAP-cAMP system by glucose
